    Double Ensemble Approaches to Predicting Firms’ Credit Rating

    Several rating agencies such as Standard & Poor\u27s (S&P), Moody\u27s and Fitch Ratings have evaluated firms’ credit rating. Since lots of fees are required by the agencies and sometimes the timely default risk of the firms is not reflected, it can be helpful for stakeholders if the credit ratings can be predicted before the agencies publish them. However, it is not easy to make an accurate prediction of credit rating since it covers a variety of range. Therefore, this study proposes two double ensemble approaches, 1) bagging-boosting and 2) boosting-bagging, to improve the prediction accuracy. To that end, we first conducted feature selection, using Chi-Square and Gain-Ratio attribute evaluators, with 3 classification algorithms (i.e., decision tree (DT), artificial neural network (ANN), and Naïve Bayesian (NB)) to select relevant features and a base classifier of ensemble models. And then, we integrated bagging and boosting methods by applying boosting method to bagging method (bagging-boosting), and bagging method to boosting method (boosting-bagging). Finally, we compared the prediction accuracy of our proposed model to benchmark models. The experimental results showed that our proposed models outperformed the benchmark models

    What Is It to Be Mentally Healthy from the North Korean Refugees' Perspective?:Qualitative Research on the Changes in Mental Health Awareness among the North Korean Refugees

    Objective: We investigated how mental health awareness among North Korean refugees transformed depending on temporal-spatial context changes. Methods: In 2013, we conducted interviews with 10 refugees (eight women) who had been in South Korea for over a year and performed a qualitative analysis of the change in mental health awareness in the differences between living in North Korea, escape (a related period of forced sojourn in a third country), and settlement in South Korea. Results: We classified 39 concepts into five main categories. The first two categories (while living in North Korea) were "a mindset for the system, but not for individual mental health" and "being confined in a social environment that was indifferent to mental health." A third category appeared during escape: "focusing on survival amid continuity of intense suffering." The final two categories appeared when settling in South Korea: "recognition of mental health amid cultural shock" and "introspection and sorting oneself out". Conclusion: This qualitative study enabled a better multi-dimensional understanding of the social and cultural aspects involved in improving mental health awareness among North Korean refugees in South Korea. It is desirable to integrate mental health as a part of daily life and to expand training for North Korean settlers

    Left dominance of EEG abnormalities in patients with transient global amnesia

    AbstractPurposeTransient global amnesia (TGA) is a syndrome of unknown etiology. Electroencephalographic (EEG) abnormalities in TGA have been reported previously. We analyzed the frequency and characteristics of EEG abnormalities in patients with TGA.MethodsWe collected EEGs of patients with a clinical diagnosis of TGA who had visited the emergency room or the outpatient clinic over a period of 8 years and compared clinical and demographic characteristics of the patients with normal EEGs with those with abnormal EEGs.ResultsEEG abnormalities were found in 35 (22.9%) out of 153 patients and epileptiform discharges were seen in 26 (74.3%) out of these 35 patients. Spikes or sharp waves were detected on the left side only (48.6%) or on both sides (25.7%), but none of the patients showed spikes or sharp waves on right side only. In six patients the EEG had normalized within three months of presentation, in ten within six months, and in twelve by one year. The EEG remained abnormal in eleven out of the 23 patients one year after presentation.ConclusionIn this largest consecutive EEG study at one center, the proportion of patients with TGA in whom epileptiform discharges were demonstrated within days of the episode of TGA was significantly higher than in the previous literature. EEG abnormalities such as spikes or sharp waves spontaneously disappeared in almost half of cases over one-year of follow-up. There was a clear left dominance of EEG abnormalities in patients with TGA

    Resveratrol induces autophagy by directly inhibiting mTOR through ATP competition

    Resveratrol (RSV) is a natural polyphenol that has a beneficial effect on health, and resveratrol-induced autophagy has been suggested to be a key process in mediating many beneficial effects of resveratrol, such as reduction of inflammation and induction of cancer cell death. Although various resveratrol targets have been suggested, the molecule that mediates resveratrol-induced autophagy remains unknown. Here, we demonstrate that resveratrol induces autophagy by directly inhibiting the mTOR-ULK1 pathway. We found that inhibition of mTOR activity and presence of ULK1 are required for autophagy induction by resveratrol. In line with this mTOR dependency, we found that resveratrol suppresses the viability of MCF7 cells but not of SW620 cells, which are mTOR inhibitor sensitive and insensitive cancer cells, respectively. We also found that resveratrol-induced cancer cell suppression occurred ULK1 dependently. For the mechanism of action of resveratrol on mTOR inhibition, we demonstrate that resveratrol directly inhibits mTOR. We found that resveratrol inhibits mTOR by docking onto the ATP-binding pocket of mTOR (i.e., it competes with ATP). We propose mTOR as a novel direct target of resveratrol, and inhibition of mTOR is necessary for autophagy inductionopen

    Loss of adipose TET proteins enhances ??-adrenergic responses and protects against obesity by epigenetic regulation of ??3-AR expression

    ??-adrenergic receptor (??-AR) signaling plays predominant roles in modulating energy expenditure by triggering lipolysis and thermogenesis in adipose tissue, thereby conferring obesity resistance. Obesity is associated with diminished ??3-adrenergic receptor (??3-AR) expression and decreased ??-adrenergic responses, but the molecular mechanism coupling nutrient overload to catecholamine resistance remains poorly defined. Ten-eleven translocation (TET) proteins are dioxygenases that alter the methylation status of DNA by oxidizing 5-methylcytosine to 5-hydroxymethylcytosine and further oxidized derivatives. Here, we show that TET proteins are pivotal epigenetic suppressors of ??3-AR expression in adipocytes, thereby attenuating the responsiveness to ??-adrenergic stimulation. Deletion of all three Tet genes in adipocytes led to increased ??3-AR expression and thereby enhanced the downstream ??-adrenergic responses, including lipolysis, thermogenic gene induction, oxidative metabolism, and fat browning in vitro and in vivo. In mouse adipose tissues, Tet expression was elevated after mice ate a high-fat diet. Mice with adipose-specific ablation of all TET proteins maintained higher levels of ??3-AR in both white and brown adipose tissues and remained sensitive to ??-AR stimuli under high-fat diet challenge, leading to augmented energy expenditure and decreased fat accumulation. Consequently, they exhibited improved cold tolerance and were substantially protected from diet-induced obesity, inflammation, and metabolic complications, including insulin resistance and hyperlipidemia. Mechanistically, TET proteins directly repressed ??3-AR transcription, mainly in an enzymatic activity-independent manner, and involved the recruitment of histone deacetylases to increase deacetylation of its promoter. Thus, the TET-histone deacetylase-??3-AR axis could be targeted to treat obesity and related metabolic diseases

    Detection of Diabetic Eye Disease from Retinal Images Using a Deep Learning Based CenterNet Model

    Diabetic retinopathy (DR) is an eye disease that alters the blood vessels of a person suffering from diabetes. Diabetic macular edema (DME) occurs when DR affects the macula, which causes fluid accumulation in the macula. Efficient screening systems require experts to manually analyze images to recognize diseases. However, due to the challenging nature of the screening method and lack of trained human resources, devising effective screening-oriented treatment is an expensive task. Automated systems are trying to cope with these challenges; however, these methods do not generalize well to multiple diseases and real-world scenarios. To solve the aforementioned issues, we propose a new method comprising two main steps. The first involves dataset preparation and feature extraction and the other relates to improving a custom deep learning based CenterNet model trained for eye disease classification. Initially, we generate annotations for suspected samples to locate the precise region of interest, while the other part of the proposed solution trains the Center Net model over annotated images. Specifically, we use DenseNet-100 as a feature extraction method on which the one-stage detector, CenterNet, is employed to localize and classify the disease lesions. We evaluated our method over challenging datasets, namely, APTOS-2019 and IDRiD, and attained average accuracy of 97.93% and 98.10%, respectively. We also performed cross-dataset validation with benchmark EYEPACS and Diaretdb1 datasets. Both qualitative and quantitative results demonstrate that our proposed approach outperforms state-of-the-art methods due to more effective localization power of CenterNet, as it can easily recognize small lesions and deal with over-fitted training data. Our proposed framework is proficient in correctly locating and classifying disease lesions. In comparison to existing DR and DME classification approaches, our method can extract representative key points from low-intensity and noisy images and accurately classify them. Hence our approach can play an important role in automated detection and recognition of DR and DME lesions

    CRISPR/Cas9-induced knockout and knock-in mutations in Chlamydomonas reinhardtii

    Genome editing is crucial for genetic engineering of organisms for improved traits, particularly in microalgae due to the urgent necessity for the next generation biofuel production. The most advanced CRISPR/Cas9 system is simple, efficient and accurate in some organisms; however, it has proven extremely difficult in microalgae including the model alga Chlamydomonas. We solved this problem by delivering Cas9 ribonucleoproteins (RNPs) comprising the Cas9 protein and sgRNAs to avoid cytotoxicity and off-targeting associated with vector-driven expression of Cas9. We obtained CRISPR/Cas9-induced mutations at three loci including MAA7, CpSRP43 and ChlM, and targeted mutagenic efficiency was improved up to 100 fold compared to the first report of transgenic Cas9-induced mutagenesis. Interestingly, we found that unrelated vectors used for the selection purpose were predominantly integrated at the Cas9 cut site, indicative of NHEJ-mediated knock-in events. As expected with Cas9 RNPs, no off-targeting was found in one of the mutagenic screens. In conclusion, we improved the knockout efficiency by using Cas9 RNPs, which opens great opportunities not only for biological research but also industrial applications in Chlamydomonas and other microalgae.
